Educational objectives Acquisition by the student of autonomy and familiarity in the use of some of the main methods applied in the field of biological investigations. Ability to identify and critically review the most appropriate techniques for achieving a given experimental objective. Critically evaluate advantages and disadvantages in the use of certain methodologies also on the basis of their sensitivity, specificity and accuracy.

Prerequisites nothing
Teaching methods he practical activity in the laboratory includes a short introductory lecture in which the experimental protocol to be followed by all students will be provided and explained. The students will conduct the experiments individually or in small groups and, at the end, the experimental data obtained will be subject to comparative analysis and discussion.

Learning verification modality There will be 5 laboratory exercises in the topics listed below for a total of 24 hours. In order to obtain the credits envisaged for this teaching (2 CFU), attendance at at least 4 of the 5 experiences is compulsory. Students' attendance will be ascertained by roll call and acquisition of signatures. Passing is subject to an aptitude test.
Extended program The course includes five laboratory experiences in which different protocols relating to some of the main methodologies used in various fields of biological experiments will be applied, with particular reference to the contents of the third-year teaching. Experiences will be carried out concerning:
1. PHYSIOLOGY: Preparation of cultured cells for electrophysiology studies. Recording of ionic currents through membrane channels. Data analysis and inferences on the kinetics of ion channels.
2. VEGETABLE PHYSIOLOGY: Extraction and quantification of photosynthetic pigments and analysis of their absorption spectra. Isolation of thylakoidal membranes and measurement of the rate of photosynthetic electron transport.
3. HYGIENE: : Sampling techniques of water intended for human consumption and surface waters and analysis of some microbiological parameters with reference to the judgement of potability and quality classification according to the provisions of D.Lgs 18/2023 and D.Lgs 152/2006 respectively.
4. CHEMICAL AND QUALITY OF FOODS Use of high-performance liquid chromatography (HPLC) in food quality control. Determination of the main chromatographic parameters; quali-quantitative analysis of bioactive components extracted from food matrices.
5. MOLECULAR BIOLOGY: Recombinant DNA technology: Methods of nucleic acid extraction and analysis. Cloning and selection of recombinant DNA.
