Contents Prevention and safety in the chemistry laboratory. Basic knowledge on separation and purification techniques of chemical compounds: filtration, extraction, recrystallization, distillation, and chromatography. Determinations of acidity/basicity constants by acid-base titration. Practical exercises in the laboratory are planned.

Educational objectives The fundamental techniques for the purification and separation of organic compounds and the principles that are the basis of these techniques will be learned along with the determination of the concentration of solutions by volumetric analysis. By the end of the course, the student should be able to prepare solutions at specific concentration, to determine the concentration of unknown solutions, to perform a filtration, and to perform a thin-layer chromatography and a column chromatography.
Prerequisites In order to understand the given topics, a good knowledge of the basic concepts of general chemistry, on the chemical bonding, acidity and basicity, equilibrium constants, is required.
Teaching methods The course is organized as follows:

- Lectures on all subjects of the course (7 h)

- Practical laboratory exercises.
Students will be divided in different groups (20-30 students per group maximum) and will perform four lab experiences (3 h each):
1) Preparation of solutions with a defined concentration;
2) Determination of the concentration of an acetic acid solution by the volumetric acid-base titration;
3) Synthesis of phenyl benzoate: work-up of the reaction and filtration
4) Characterization of the synthesized phenyl benzoate by TLC analysis and melting point.

Learning verification modality There is not a final examination. However, at the end of each practical lab experience a laboratory report should be filled.
In order to acquire the CFU, students will have to attend at least 75% of laboratory exercises. Frequency will be verified by roll-call and signature take-over.
Extended program Prevention and safety in the chemistry laboratory. Separation and purification techniques of chemical compounds. Filtration: simple and under vacuum, vacuum pumps. Extraction: the partition coefficient, selective extraction, discontinuous and continuous extraction. Recrystallization and melting point. Distillation: simple and fractional. Chromatography: theoretical principles, thin layer chromatography, column chromatography, gas chromatography and HPLC. Determination of acidity/basicity constants by acid-base titration. Volumetric analysis. pH indicators.

Practical exercises in the laboratory are planned regarding the preparation of solutions with defined concentration, the acid-base titration, the thin layer chromatography and column chromatography.
